Since it’s System holding it, I imagine you’ve opened up the ISO which mounted the image as a drive letter, in which case you can clear the problem by unmounting the ISO. You can do that by opening up File Explorer, right-clicking the mounted drive and choosing Eject.
Although you probably don’t need it for this situation, I like to use LockHunter for solving the general locked files problems that arise from time to time.
